Just booked 6 days away at the end of May. It's a first-time visit for me. I have guide books from the library but interested if anyone has any particular recommendations of must (or must not!)things to see or do.

We had 4 great days there a few weeks back. Take comfy shoes - there's plenty of walking to be done. If you haven't book accommodation yet we had an excellent apartment in the very centre which was good (email in profile)
Typical tourist-y things like the castle views etc are good on a sunny day. Don't book tickets for the first concert you see advertised - there are dozens all over town. Stick to the small narrow streets and follow your nose around.
Eating out in the centre is not cheap and (in our experience nothing special). Beer is good as one might expect. Stick to 'AAA' taxi cabs with meters - some of the others end up being much more expensive. Take plenty cash, CC's are not welcome in all places (most but not all).
But ignoring all the hints it is a great place and I'm sure you'll have a great time.

What IanG said - it's a great place to just walk around.
With 6 days, you may well want to get out of the city and take a trip to elsewhere - Cesky Krumlov is a bit of a trek but worth it.
One word of warning - we found the hard way that they eat early in Prague! And if you're vegetarian... well it's not impossible but it is a bit tricky.

I guess it's a case of....
"Do you have anything for vegetarians?"
"Yes. Contempt"
Luckily I'm only a pretend vegetarian, so I should be ok.
I have a very reasonably-priced and nice-looking apartment a little out of the centre very near the castle. The company SEEM very efficient and friendly as well. I'm very much looking forward to it.
Thanks for the comments so far.

"The Good Soldier Svejk" by Jaroslav Hasek was one of the first (if not the very first) war books with an anti-hero and is a Czech institution. Part was written in Hostinek U Kalich, a hotel near I.P.Pavlova metro station. If you go to the pub section (left hand side, facing the hotel), the food is good traditional fare, the beer is excellent and the staff welcoming (at least, when I was there last). Just ask for the "Svejk Restaurant".
